Monocytes are the population that most often makes a PBMC preparation disappoint, and the failure is rarely obvious while it is happening. They are fragile relative to lymphocytes, they respond to handling by activating, and they adhere to almost everything. A protocol written without accounting for those three properties will produce a result, and the result will be about the handling.
What CD14 identifies
CD14 is a surface receptor that is part of the machinery recognising bacterial lipopolysaccharide, and it is expressed most strongly on classical monocytes. Selecting on it captures the dominant monocyte population cleanly.
The nuance is that monocytes are not homogeneous. Classical monocytes express CD14 strongly with little CD16, non-classical monocytes carry the reverse pattern, and an intermediate population expresses both. CD14 selection enriches for the classical subset, which is usually what is wanted and is occasionally not.
The fragility problem
Monocytes tolerate cryopreservation poorly compared with lymphocytes. Recovery after thaw is routinely lower than their frequency in the input material, which means a thawed PBMC preparation is depleted of monocytes relative to the fresh preparation it came from.
This matters twice. It skews subset ratios in any analysis assuming input frequencies, and it undermines any downstream workflow depending on monocyte number, most obviously monocyte-derived dendritic cell or macrophage generation. They activate when handled
Monocytes are sentinel cells, built to detect and respond. Adherence to plastic activates them. Prolonged handling activates them. Endotoxin contamination activates them strongly, which is why endotoxin on a certificate of analysis matters more for monocyte work than for most other applications.
The practical consequence is that a monocyte preparation is rarely in a truly resting state by the time it is assayed, and the degree of pre-activation varies with handling. Recording handling time and including an activation baseline turns that from an unknown into a measured variable.

For work where resting state matters, negative selection or elutriation is worth the additional input. For work where the monocytes will be deliberately activated anyway, the simpler methods are defensible.
What they are used for
Monocyte-derived dendritic cells and macrophages are the largest use, generated by differentiation in culture over several days. Beyond that, monocytes appear in innate immunity work, inflammation models, phagocytosis and cytokine response assays, and increasingly in engineered myeloid cell approaches.
Macrophages themselves are generally produced by differentiating monocytes rather than isolated directly, which is why the quality of the monocyte input determines the quality of everything downstream of it.
